I found some noticeable new features right away with WordPress 2.8, including the editor which contains numbered lines now in addition to color coding the html tags of your php files. This makes it much easier for making manual HTTP edits of your php files so you can add in code to your sidebar.php or manipulate your header.php.

Unfortunately one thing now I have noticed on all 4 of my blogs that I have upgraded is when creating a “new post” there can be stickiness when clicking between the visual and HTML tabs, sometimes you can’t click on one tab or the other, it just doesn’t work unless you go to a different page and come back. Other times you click on HTML editor and you are stuck on Visual editor instead.
I have noticed this with all four of my blogs and have never seen this issue prior to WordPress 2.8 (2.7.1 never presented this problem), it is a cosmetic issue, but it is annoying to have to leave my post and come back to it to switch the editor tab. This happens to me quite often and I notice it is worse if you try to switch tabs too quickly.
The New Post could still use some enhancements like page line numbers (just like the editor) and some more text formatting options, but I like that underline has been added (not sure if that was with 2.7 or 2.8, I just noticed it recently). One thing that would be a nice feature is if the editor mentioned somewhere the “width” set in your php for your “post” area, so you knew exactly how big your post area was, since I run 4 blogs I am constantly confusing my post area size. It would be nice to have just a little display field showing you the set “width” of your post field, so you can always size images properly and such.
In the meantime all of my plugins stayed intact and none of them failed as a result of upgrading from WordPress 2.7.1 to 2.8 so that is a good thing.
